Modulation of Induced Cytotoxicity of Doxorubicin by Using Apoferritin and Liposomal Cages

Doxorubicin is an effective chemotherapeutic drug, however, its toxicity is a significant limitation in therapy. Encapsulation of doxorubicin inside liposomes or ferritin cages decreases cardiotoxicity while maintaining anticancer potency. We synthesized novel apoferritin-and liposome-encapsulated forms of doxorubicin ("Apodox" and "lip-8-dox") and compared its toxicity with doxorubicin and Myocet on prostate cell lines. Three different prostatic cell lines PNT1A, 22Rv1, and LNCaP were chosen. The toxicity of the modified doxorubicin forms was compared to conventional doxorubicin using the MTT assay, real-time cell impedance-based cell growth method (RTCA), and flow cytometry. The efficiency of doxorubicin entrapment was 56% in apoferritin cages and 42% in the liposome carrier. The accuracy of the RTCA system was verified by flow-cytometric analysis of cell viability. The doxorubicin half maximal inhibition concentrations (IC50) were determined as 170.5, 234.0, and 169.0 nM for PNT1A, 22Rv1, and LNCaP, respectively by RTCA. Lip8-dox is less toxic on the non-tumor cell line PNT1A compared to doxorubicin, while still maintaining the toxicity to tumorous cell lines similar to doxorubicin or epirubicin (IC50 = 2076.7 nM for PNT1A vs. 935.3 and 729.0 nM for 22Rv1 and LNCaP). Apodox IC50 was determined as follows: 603.1, 1344.2, and 931.2 nM for PNT1A, 22Rv1, and LNCaP

Tato práce se zabývá bezpečností protokolu IPv6 se zaměřením na lokální sítě (LAN) a IDS/IPS systémy. Porovnává zranitelnosti a hrozby protokolů IPv4 a IPv6 včetně základních bezpečnostních doporučení. Vybrané útoky související s IPv6 jsou pak demonstrovány v simulované situaci útočník/oběť na síti s protokolem IPv6. Tyto experimenty jsou pak použity k zhodnocení použitelnosti IDS/IPS zařízení proti hrozbám specifickým pro prostředí protokolu IPv6.Katedra informatiky a výpočetní technikyObhájenoThis thesis reviews IPv6 security with focus on Local Area Networks and IDS/IPS systems. It compares IPv4 and IPv6 threats, vulnerabilities and gives basic security recommendations. Selected IPv6 attacks and exploits are demonstrated in simulated attacker/victim scenario on IPv6 network. These experiments are then used to set up guidelines for evaluating usability of IDS/IPS appliances for IPv6-specific threats

Effect of CNC Interpolator Parameter Settings on Toolpath Precision and Quality in Corner Neighborhoods

Surface quality, machining time, and precision of the final workpiece are key criteria of optimization in CNC machining. These criteria are influenced by multiple factors, such as path interpolation, feed drive system settings, machine dynamics, and the manufacturing process. The properties of the output of the interpolator indirectly influence all subsequent phases of the machining process, thus influencing the quality of the end product. This paper focuses on the effects of interpolator settings on toolpath quality and precision in corner neighborhoods for the commercial Heidenhain iTNC interpolator. A novel method of toolpath quality evaluation suitable for interpolator output toolpaths is proposed, and the effect of multiple CNC parameters on toolpath quality and precision in corner neighborhoods is quantified based on results obtained on a testing toolpath and verified on a toolpath composed of linear segments only. Both toolpath quality and precision were found to depend primarily on the parameters of limit frequency, contour tolerance, and corner jerk settings with precision additionally depending on angle size. The results show that both toolpath quality and precision in corner neighborhoods can be successfully controlled by the corner jerk limit parameter settings. The presented methodology provides a practical guide for CNC parameter settings in Heidenhain interpolators aimed at predicting toolpath quality and precision in corner neighborhoods

Smooth Acceleration of Transverse Axis Movements in CNC Threading Machining

Most of the complex details of modern technical products are manufactured on CNC equipment. The threaded surface is one of the most critical elements of such parts. Their reliability and durability largely depend on the accuracy of the machining process. Machining threaded surfaces requires precise synchronization of periodic tool movements with the continuous rotation of the spindle. Ensuring smooth movement along the transverse axis with a synchronized approach to the machined surface is one of the important tasks. A variant of solving this problem in a two-level CNC system is proposed. A mathematical model of the movement along the ransverse axis with a smooth variation in acceleration has been developed. A technique is proposed using an S-shaped feed profile based on the sin² function, which provides ease of calculation and high accuracy. The model is implemented in a two-level CNC. The results of experimental studies are shown that servo error decreases to a value of the order of 0…–1 μm before the end of the approach section, outside the workpiece

An optical trap apparatus to study the thermophoretic phenomena

    No full text
The work is aimed at the development of the experimental device to study of the motion of different colloidal objects in the combined force field composed of the optical-trapping force originating in the light-intensity gradient and the thermophoretic force caused by the temperature gradient in aqueous solvents. Polystyrene microscopic beads of sub-micrometer diameters are used as a simple model of the optical tweezers calibration. The methods of generating temperature gradient are under development. A dual optical trap is used as a tool for the investigation of the thermophoretic force acting on the observed particles. Also, the generated optical trap's force constant is calculated by various methods, including Fast Fourier Transform, and the influence of differences in the sample environment and light intensity is compared. Results of these experiments should contribute to the theory of the optical trapping effect, so far not understood in detail, and should open a way for developing novel methods of the investigation of thermophoretic phenomena in a water environment. © 2022 SPIE.CZ.02.2.69/0.0/0.0/19_073/001694
